报告题目:Fluorescent organic nanoparticles and advanced fluorescence imaging techniques: design and applications

报告摘要:

The talk will be divided in two parts. The first part will be devoted to the design and applications of  fluorescent organic nanoparticles (NPs) with high brightness. We notably designed stable nano-emulsion droplets encapsulating lipophilic fluorescent derivatives at exceptionally high concentrations. These highly fluorescent nano-droplets were used to image the blood flow in zebrafishs. We also designed fluorescent polymer NPs brighter than QDs and exhibiting photo-induced reversible on/off fluorescence switching. This collective switching of hundreds of dyes was related to ultrafast excitation energy transfer and could be used for super-resolution imaging. Last, we designed efficiently loaded and stable sub-20-nm polymer particles that preserved the capacity to encapsulate large amounts of ionic dyes, generating NPs 10-fold brighter than QDs of the same size. The second part will deal with advanced fluorescence microscopy techniques that were implemented in the laboratory. Quantitative imaging techniques as well as high resolution techniques afford unprecedented information on the dynamics and interactions of proteins in live cells. The applications of these techniques will be illustrated with a protein of the Human immunodeficiency virus type 1 (HIV-1).

Research interests

Head of a team of 20 senior researchers and 30 students, my research activity can be divided in two research axis. 1) Development of cutting-edge fluorescence techniques (quantitative two-photon microscopy combining FLIM and FCS, single molecule spectroscopy, high resolution microscopy such as dSTORM/PALM, STED and AFM/fluorescence microscopy) and innovative environment-sensitive fluorescence probes. 2) Application of these techniques and probes to monitor biomolecular interactions, mainly with viral proteins.
